Protein-bound Dye and Purine-catabolyzing Enzymes in Parenchymal and Nonparenchymal Cells of Rats Fed 4-dimethylaminoazobenzene.

چکیده
The binding of 4-dimethylaminoazobenzene to proteins and the enzymatic activities of some purine-catabolyzing enzymes during feeding of the carcinogen have been in vestigated by analysis of two liver fractions differing in their cytological composition. At an early stage of feeding dye binding occurred at a faster rate in nonparenchymal cells than in parenchymal cells. Binding in nonparenchymal cells reached a maximum after 1 week of feeding and decreased afterward, whereas maximum binding in the parenchymal cells was observed after 4 weeks. The decrease in the amount of bound dye in parenchymal cell that followed was accompanied by a second peak in nonparen chymal cells. The results on the activities of the purine-catabolyzing enzymes show that a de crease in 5'-nucleotidase, inosine phosphorylase, and uricase activities occurred in

Studies on the intracellular composition of liver and liver tumor from rats fed 4-dimethylaminoazobenzene.
In a previous study (8) the livers of rats fed the hepatic carcinogen, 4-dimethylaminoazobenzene, were separated by differential centrifugation into the nuclei, large granules (mitochondria), small granules (microsomes), and a supernatant fluid (particles nonsedimentable at 19,000 X g). Anal yses for protein, nucleic acids, riboflavin, and pro tein-bound aminoazo dye were made on each frac tion...
متن کاملRetinol and retinyl esters in parenchymal and nonparenchymal rat liver cell fractions after long-term administration of ethanol.
Chronic ethanol consumption reduces the liver retinoid store in man and rat. We have studied the effect of ethanol on some aspects of retinoid metabolism in parenchymal and nonparenchymal liver cells. Rats fed 36% of total energy intake as ethanol for 5-6 weeks had the liver retinoid concentration reduced to about one-third, as compared to pair-fed controls. The reduction in liver retinoid affe...

INTRODUCTION Miller and Miller (3) showed that the carcino genic dye, 4-dimethylaminoazobenzene, is firmly bound to protein constituents of the rat liver. Such protein-binding reaches a maximum after 4-6 weeks on the azo diet, whereas the neoplasm arises several weeks later. Moreover, the neoplastic tis sue contains essentially no protein-bound dye. Hence, Miller and Miller (3) have postulated ...
متن کاملCertain effects of dietary fats on the production of liver tumors in rats fed p-dimethylaminoazobenzene.
Hydrogenated coconut oil and its component fatty acids appear to be the most striking examples of dietary lipid inhibitors reported for the carcinogen p-dimethylaminoazobenzene. When these substances were fed to rats as the sole sources of lipid in synthetic diets containing the azo dye very few liver tumors, if any, appeared by 6 months (3, 4). If," however, a fat such as corn oil was fed inst...
متن کاملThe in vitro formation of protein-bound derivatives of aminoazo dyes by rat liver preparations.
Previous studies have demonstrated the presence of protein-bound aminoazo dyes in the livers of rats fed the hepatocarcinogen 4-dimethylaminoazobenzene (DAB) 1 or certain of its derivatives ( ~ , 35). Since, under a variety of conditions, the levels of these bound derivatives can be correlated with the expected tumor incidence, the formation of the protein-bound dyes appears to be an important ...
